The Trousers FAQ is a great source of TPM information.

Amit Singh's page is also packed with information (the page is geared towards setting up your TPM on the Mac so you'll need to read between the lines).

Of course the TPM Work Group section of the Trusted Computing Group contains required reading. Their FAQ is of minor interest, but you'll also find the latest spec (1.2) in the Specs section, and header files here, along with TSS information.
